Tumor Marker PSA Test
The INTEX One-Step PSA Test is an in-vitro immunochromatographic one-step assay designed for a qualitative and a semi-quantitative determination of human prostate specific antigen (PSA) in serum.

Clinically, prostate cancer is now the most frequently diagnosed cancer for males and the second most common cause for mortality due to cancer. Its incidence increases dramatically in males over 40 years of age, occurring in 50% of those over 70 years of age (1,2,3). Compared to other cancers, prostate cancer is more successfully treated if diagnosed early.

Digital rectal examination has been a useful procedure in early detection of prostate cancer but has its limitations (4). Also, the biochemical markers which are available to monitor this disease (i.e., prostatic acid phosphatase, total alkaline phosphatase, carcinoembryonic antigen, creatine kinase isoenzyme BB, etc.) are not specificly made for prostate cancer. Therefore, the management of patients with prostate cancer has been hindered by the lack of a more accurate markers.

Recently, another prostate enzyme has been identified and purified, that is specific for prostate tissue, normal or malignant and also found in periureteral glands. It is called prostate specific antigen (PSA) (6,7). Human PSA is a single chain glycoprotein with a molecular weight of approximately 34,000 dalton containing 7% carbohydrate by weight. Immunohistochemical studies have shown that PSA is confined to the cytoplasm of prostatic acinar cells and ductal epithelium. PSA is present in normal, benign hyperplastic, and malignant prostatic tissue, in carcinoma, and also prostatic fluid and seminal plasma. PSA is not present in any other normal tissue obtained from men, nor is it produced by cancers of the breast, lung, colon, rectum, stomach, pancreas and thyroid. Purified PSA lacks any prostate acid phosphatase (PAP) and vice versa. Therefore it is biologically and immunologically distinct from PAP. PSA has been prooven to exist with two forms: free PSA and PSA complexed with the serine prostate inhibitor anti-chymotrypsin.

Elevated levels of PSA in serum have been detected in men with prostate cancer, benign prostate hyperplasia (BPH) and in some cases of inflammatory disorders of other genitourinary tissues in proximity to the prostate (12,13). Increased levels are not observed in healthy men or those with cancers other than the prostate. Females who are healthy or have cancer do not exhibit PSA. Serial testing for elevated levels of PSA in serum can provide an important tool in the monitoring of disease recurrence or in following the effectiveness of a particular treatment.
